Options!
One of the biggest challenges faced by game developers is how to appeal to a wider audience.
Often any choice they make will appeal to one side of the demographic and alienate another.
But by introducing more options each gamer can adjust the game to their liking.
So instead of just having basic difficulty settings like easy, moderate and hardcore, which can be there too, you can allow the player to expand on that if they chose to. So things like marking targets, highlighting pickupable items, features to be included on map and hud etc should all be up to the player to change. Basically if you are thinking "should we include this feature or not?", include it always but make it optional (if it is doable, of course, some features cannot be optional). This is especially helpful if the devs have ALREADY decided to include the feature but are not 100% sure it will be popular. Just make it optional.
I for one love to be able to do orienteering when playing sandbox games and Far Cry 3 would have been a MUCH better experience for me if only I did not see the "you are here" arrow on the map screen. You can see where you are by the land features around you and if you cannot immediately than you can move to some high ground and check again. This would have been, I imagine, VERY easy and cheap to implement with a single option in the menu.
But i digress. I think that a game would lose nothing by having more options and instead satisfy more discerning players, thus broadening its appeal.
If you are concerned about making it too complicated make sure you hide the options under an "advanced options" tab and, for goodness sake, provide proper descriptions of what each option actually does (SO many games have explanations of features or options appear when you hover the cursor over them only to repeat verbatim what the descriptor said in the first place). A well layed out options screen that is not compulsory for casual players to go through (they can just chose between easy, medium and hard) would add A LOT to the game's popularity.
This is, I think, the best way by which you can appeal to both the casual and more mature market.
Inventory system such as the one in the STALKER series (has it been mentioned already?).
Basically having limited space and/or weight that can be expanded depending on whether or not one has a backpack, ammo belt, holsters etc.
Different sizes and types of holding equipment can be implemented depending on the dev's choosing.
Different difficulty levels would vary the limits imposed, with the hardcore mode imposing more realistic values.
As an option a penalty may be imposed on overloading the character in mobility, stealth level or health drop

NECESSARY ADDITIONS:
They need to have a server browser just like they did in FC2 and FCIP, FC3 was an absolute nightmare to find a good game and get a lobby started in.
Also the inconsistency between the map editor physics/visuals and the in game multiplayer physics/visuals was really annoying. A good example would be making a jump that you could just make in the editor but when you try the same jump in a multiplayer game you wouldn't be able to make it.
The map rating system is stupid and should be removed all together or at most it should just be done on the back-end, if a map is good people will play it (assuming it's easy to find a match!).
Put vehicles back into the multiplayer, I really never understood the whole point of taking them out?
Add a CTF/STS game mode where we can throw the objective like in FCIP, that added a really cool dynamic to the game mode and it was A LOT of fun.
ADDITIONS THAT WOULD BE COOL BUT NOT NECESSARY:
Allow us to put AI into our multiplayer maps, nothing would be cooler than being in a gunfight and suddenly a tiger mauls the guy you are shooting at!
Character customization similar to FCIP, I did not like the character models in FC3 so it would have been cool to change the clothes and gear to my liking.
Triggers/events like in the Trials editor, it would AWESOME if I could trigger a rock slide or something that crushes a group of people going for the objective!

Hey A_Combi_Stick - here is a FC4 Wish
If we get an editor and some fun game modes than put together an Orthodox style map especially if we get a Solo Deathmatch type mode.
That was a killer of a map.
I really hope we get some good fog - FCIP was excellent for fog and no other FC game came close on consoles, getting some snow storm effects / thin breakable ice for floors and walls could be interesting to drop into or shoot into shortcuts in a map.
