I think after the story of Aguilar and Maria, we could move on to Ezio's ongoing story and Italy as a New Region would be great.
Some non-added heroes from AC Brotherhood and AC2 could be added to the game: Leonardo da Vinci, Rosa (Venetian Thief), Caterina Sforza and others.
We can have different versions of Ezio, Ezio in "AC Brotherhood" and new gear, new skills, and then Ezio in "Revelations" and new gear, new skills.

what I like best of your suggestions is the idea in general to extend the story, not the variety of events. Continuing in Spain is one possibility. For Italy they should create a second map, I think, with new chapters/regions beginning at 1. The next could be Greece, Egypt, Levante, Carribean etc.
Some missions could contain a choice of characters in opposite to some events, which means that the main characters (like Altaïr, Al Mualim and Kenway) are not excluded, but explicitely required.
Introducing a 6th chapter bears the risk that the difficulty could be increased, too, so they would have to raise the characters' level to 60. No thanks. The new missions should have a really innovative aspect.
Personally, i think that there are 3 most optimal options for a new main story regions:
1) Сolonial America between AC:BF-AC:III/Rogue- new toons can be based on ACBF-ACIII multiplayer characters. That option can introduce Connor, Shay, etc. as grindable characters in legacy missions (so that will guarantee that they won't become PayConnor and PayShay).
2) Сontinuing Aguilar's story with sending him to a New World with Columbus (and apple of eden). So new location here is a New World.
3) A lot of new regions that will show us stories of assassins from AС:Reflections and AC:Project legacy
