How can I unlock the locked content on my Rocksmith 2014?

you mean Karawan? You have to link your Uplay account to Rocksmith

You also can do 250 missions or something like that.

I lost my entire Rocksmith steam profile in May and had to start over from scratch.

It took me about 23 days to get 250 missions. The only thing I wanted was the MAX Venue size.
