Since Tier 1 operators basically have a licence to kill, assassination missions would be a welcome change of pace, which could open up a wide range of new weapons and gameplay features. For example, if my target is surrounded by a mountain of security, then disguising myself as the enemy would be the best option. If they are going to give the player the option to dress up like a ret@rd in brightly colored tights, then I should also be given the option to dress up as the enemy itself. Maybe I decide that making his death look accidental is the best approach to go, in which I would have a wide variety of weaponry that can kill without a trace, like poison dart guns that can induce a heart attack, which is a real weapon as I show in the video below.

Perhaps killing him this way was the best way I can get closer to another related target, perhaps at his funeral, so I set up a sniper point and put a bullet square in his dome.
You see on the White Hat mission that you can assassinate him, or steal his drugs and leave him to it. That essential gives you the choice to potentially turn any mission into an assassination, although I would have thought some scenarios would require a target to be taken alive to gather Intel as per the E3 2016 demo.
You would think that a top secret CIA unit would have much better means in taking him alive than just a sleeper hold. I want to see some the high tech weapons and gadgets that we know they would have. All the missions need to be interconnected with each other for all of these options to actually mean something. Pretty sure what you're advocating for (contextual, situational reactions to player actions) is one of the big selling points of this game. The White Hat mission referenced above would be an example of that, but I imagine you could do the exact same things with El Pozolero too. It's probably always the better option to take someone alive, but then again killing someone is effective too. You might not get anything out of El Pozolero by taking him out, but the cartel won't be using his services ever again. Additionally, if like to point out that when I played the E3 demo I was the one to capture the manager with the intel we were seeking, and once I was done interrogating him I killed him.
I think Hitman levels of target assassination might be a bit *too* much for a game trying to be a tactical shooter, but being able to fool guards with disguises would be very cool and a unique way to get behind enemy defenses without shooting or sneaking your way in. What I want to see though are the less-lethal options for taking targets alive like tranquilizer type weapons or tazers.

Absolutely! Even something as simple as throwing rocks ala Far Cry or Sniper Elite would be acceptable too. There has to be a way to lure guards to more convenient locations or to break up their patrol routes. I firmly believe also that moving corpses is absolutely essential too.
Yeah, not so sure on this one. I kind of ended up longing for a big wheel barrow to cart bodies around on Splinter Cell. It got so tedious lugging them into cover, rinse repeat. Taking live prisoners and moving to a hidden area and incapacitating them would be better for me. Well, as someone that has recently really gotten into stealth oriented games it's kind of essential to me. I mean, it's alright to leave bodies if they are in isolated areas but sometimes enemies just don't go where you need them to. It's sort of a risk/reward thing. Being able to move bodies also allows you to set up traps - place a body in a spot where they'll be noticed and rig them with mines or command detonated explosives. These are the kind of devilish things you can come up with playing games like Far Cry, Sniper Elite, or even Metal Gear Solid.
This is an older video when I was playing through Far Cry 4 a month or so ago where you can kind of see the importance of hiding bodies to remain undetected. Some of my language is NSFW so for people with sensitive ears be advised that I tend to curse a lot during gameplay. I didn't set any traps since I was trying for a completely stealthy run though.
